When Janet was three years old, her family relocated to Miami, Florida, after her father obtained a position with Eastern Air Lines.

Guthrie obtained his pilot’s license when she was 17 years old. She worked as a research and development engineer for an aviation corporation for six years after graduating from the University of Michigan in 1960.

She was one of four women to qualify for NASA’s scientist-astronaut program at that time, but she was later rejected when a doctorate became a requirement. She was a sports car aficionado since 1962, and she began racing frequently in 1963. She was a member of an all-female racing team from 1966 to 1971.

She originally attempted to compete in the Indianapolis 500 in 1976 but failed to qualify. Guthrie finished 15th in the 1976 World 600, becoming the first woman to compete in a National Association for Stock Car Auto Racing (NASCAR) Winston Cup superspeedway race

She qualified for the 1977 Indianapolis 500 through speed trials and became the first woman to compete in the race on May 29. However, mechanical issues caused her to quit the race after 27 laps.

Despite a damaged wrist, she qualified again in 1978 and finished for the first time, finishing ninth in the field. She also competed in the 1979 Indianapolis 500, but due to mechanical issues, she was unable to finish. On April 27, 2006, she was inducted into the International Motorsports Hall of Fame.
